SUMMARY:
Dengue, a viral infection prevalent in tropical regions, shows local variations in risk influenced by
climatic, socioeconomic, and environmental factors. This study aims to identify the dengue virus
serotypes responsible for the 2022 outbreak in Nepal, where limited molecular information on serotypes
is available. Serum samples from suspected dengue patients visiting Dhulikhel Hospital were analyzed
using serological and molecular tests. Out of 89 positive samples, RNA was extracted and subjected to
RT-PCR for serotype identification. Results showed a higher seropositivity rate in females (52%)
compared to males. Positive cases were distributed across 14 districts, with the highest percentage in
Kavre (58.4%). Molecular investigation revealed that DENV1 was the predominant serotype, followed
by DEN3 (24.5%) and DENV2 (16.9%), with no detection of DENV4. The average Ct value of all
serotypes was 17.6, with DENV3 having the lowest Ct value (16.6), indicating high viremia. This study
provides valuable molecular insights into the circulating serotypes of dengue virus during the 2022
outbreak in Nepal, highlighting the prevalence of DENV1 and the association of DENV3 with high viral
load.
